Resveratrol increases the sensitivity of multiple myeloma cells against bortezomib via Hedgehog signaling pathway

Purpose: To investigate the effect of resveratrol (RSV) on bortezomib (BTZ)-resistant multiple myeloma (MM) cells, and to elucidate underlying mechanism action.
 Methods: H929 cell lines were exposed BTZ for 8 months establish BTZ-resistant MM model. Cell proliferation was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Apoptosis measured using annexin V/propidium iodide (PI) staining while cycle analysis evaluated flow cytometry. The expression Hedgehog (Hh) signaling proteins (sonic hedgehog (SHH), smoothened (SMO), glioma-associated oncogene homolog (GLI)) analyzed western blot.
 Results: H929R confirmed as a line that is resistant BTZ. RSV enhanced sensitivity cells against via inhibition viability colony formation, induction apoptosis regulation apoptosis-related proteins. Furthermore, inhibited Hh (p < 0.05.
 Conclusion: enhances BTZ, partly pathway. Thus, pathway probable target treatment, has potentials use in clinical management MM.
